Will deleting ebooks in my download folder delete the books completely?

Hi there! I have a ton of books downloaded to my Kindle App on my computer. However, as I went through my downloads folder to try to clear some stuff, I noticed there were slots for those books in there. I was wondering, if I delete these from my folder, will it also delete them from my Kindle?
